초록 | β-amino-α-hydroxy acids have been attracted much attention since they are not only useful building blocks in many biologically active natural products but also useful chiral auxiliaries. In our previous reports, stereoselective conjugate addition of the N-hydroxymethyl group moiety in an intramolecular manner was reported to produce trans-oxazolidine-5-carboxylates which are synthons in the synthesis of β-amino-α-hydroxy acids In this study, large scale one-pot synthesis of methyl trans-oxazolidine-5-carboxylates from N-hydroxymethyl-L-phenylalaninal is introduced. During the process, phenylsulfonylnitromethane was used to induce the intramolecular conjugate addition reaction, and Nef reaction by ozonolysis gave the methyl trans-oxazolidine-5-carboxylates in 41~48% yields. |
