제목 Synthesis of PEG-b-Poly(L-lysine-co-L-Phenylalanine)s Conjugated with L-Arginine as a Non-viral Gene Delivery Vector
초록 Natural and synthetic polycations displaying non-pathogenic and non-immunogenic properties have been employed as non-viral vectors for gene delivery due to the safety concerns. The main obstacle in gene therapy is the development of efficient, non-toxic delivery carriers, which effectively transport DNA inside the cell nucleus of the target. Lysine based amino acid copolymers containing 30 molar % of phenylalanine residues (PK7F3) were prepared with α-methoxy-ω-aminopoly(ethylene glycol) block at one end. Primary amino groups in the side chain of lysine residues were conjugated with L-arginine in almost quantitative yields. Obtained copolymers were applied as a gene delivery carrier and the selective targeted delivery to the neuron cells were tested with the introduction of a neuron cell specific ligands by physical mixing with the copolymers.
