제목 PEG-b-Poly(L-lysine-co-L-Phenylalanine)s Conjugated with targeting moieties as a Non-viral Gene Delivery Vector
초록 The main obstacle in gene therapy is the development of efficient, non-toxic delivery carriers, which effectively transport DNA inside the nucleus of the target cells. In this study, Lysine based amino acid copolymers containing 30% of phenylalanine residues were prepared with α-methoxy-ω-aminoPEG block at one end. Primary amino groups in the side chain of lysine residues were conjugated with L-arginine derivatives. Obtained copolymers were applied as a gene delivery carrier and the selective targeted delivery to the neuron cells were tested with the introduction of a neuron cell specific peptide by physical mixing the peptides with the copolymers and their transfection efficiency as well as cytotoxicity were tested. PEG-b-Poly(L-lysine-co-L-Phenylalanine)s conjugated with histidine derivatives were also prepared and their transfection efficiency as well as cytotoxicity were tested to investigate the role of the histidine derivatives as endosomal escape moieties.
