Proteins are important biologic therapeutics including antibodies, interferons, enzyme and vaccines used for the treatment of various diseases. However, owing to short half-time and fast clearance of the proteins in a body, long-term delivery of protein therapeutics pose a significant challenge. Here we present a double-layered microneedle array with swellable tips prepared by polystyrene-block-poly(acrylic acid) block copolymer, enabling mechanical interlocking with soft tissues. This swellable microneedle array achieved ~1.8 N/cm2 in adhesion strength to a fresh rat skin. By using reversible swelling/deswelling process, insulin as a protein drug was loaded into the swellable tips at ambient condition. The loaded insulin was released by passive diffusion through the swollen microneedle tips affixed on the rat skin. |
