Controlling stem cell fate and phenotype is important for development of cell-based therapeutics and tissue engineering products. Cell-matrix and/or cell-cell interactions are critical to regulate stem cell phenotype, such as cell adhesion, proliferation, and differentiation. Many studies have attempted to improve cell adhesion onto scaffolds by typically modification with ECM molecules, growth factors, or other bioactive molecules. However, little attention has been paid to control cell-cell interactions using synthetic ECMs. Stem cells require a specific microenvironment, in which interactions between stem cells and neighboring cells of other types exist. In this study, stem cells were cultured on alginate hydrogels modified with molecules that induce cell-cell interactions. These cell-instructive hydrogels were useful to enhance cell-cell interactions and to control differentiation lineage of the stem cells. |
