Efficient production of 3-hydroxypropionic acid from acetate by engineering metabolic pathway and optimizing carbon flux at malonyl-CoA in Escherichia coli |

Acetate is an abundant and potential feedstock for microbial processes where diverse value-added chemicals are produced. In this study, we produced 3-hydroxypropionic acid (3-HP) from acetate by engineered Escherichia coli. We initially introduced mcr (malonyl-CoA reductase) from Chloroflexus aurantiacus. Then, acetyl-CoA sythetase was overexpressed for improving assimilation of acetate, and iclR (glyoxylate shunt pathway repressor) was deleted for accelerating acetate consumption. Furthermore, we controlled carbon flux at malonyl-CoA node to prohibit malonyl-CoA from leaking to fatty acid synthesis cycle by adding cerulenin. Subsequently, we found that decreased carbon flux to fatty acid synthesis leads dramatic improvement of 3-HP production (3.00 g/L of 3-HP from 8.98 g/L of acetate). This result demonstrate that acetate can be utilized as a promising carbon source for microbial process and 3-HP can be produced from acetate with high yield (44.6% of the theoretical maximum yield). |
