제목 A Novel Reverse Gene Transfer for Neonatal Cardiomyocytes
초록 Cardiomyocytes are difficult to transfer the genes with standard protocols using commercial reagents unless viral vectors are used. In this study, we developed a safe and efficient technique for gene transfer and expression screening in cardiomyocytes with an atelocollagen complexed with nanoparticle and reporter gene. The nanoparticles containing reporter gene, pCMV-DsRed1, were embedded on atelocollagen coated culture plate. Cardiomyocytes were isolated from neonatal rat and seeded on the coated culture plate. The expression of red fluorescent protein (RFP) were confirmed by fluorescent microscopy and quantified by flow cytometry analysis. The DNA complexes immobilized on the collagen surface were efficiently transduced into cardiomyocytes without a significant cytotoxicity. These results suggest that this novel technique using atelocollagen and nanoparticle may be appropriate for general use for gene therapy in cardiomyocytes.
