Label-free Detection through Ligand Immobilization on Polydiacetylene-coated and SERS-encoded Beads |
초록 |
Multiplex protein detection technologies can obtain a better understanding of protein–protein interactions. However, high-throughput detection is difficult owing to the requirement for detect protein binding event using labeled secondary proteins. We have already determined that polydiacetylene-coated surface-enhanced Raman scattering-encoded beads (PDA–SERS beads) can offer an increased SERS signals and label-free detection through the PDA payer. In here, we successfully introduced a ligand immobilization method on PDA–SERS beads. The color of PDA–SERS beads is changed by upon exposure to external stress and SERS signals could be utilized for identification, indicating their potential for multiplex analyses. By using biotin-conjugated PDA-SERS beads, fluorescence properties were altered by specific binding of streptavidin concentration. In addition, we could detect 2ⅹ10-8 M of streptavidin using the fluorescence intensity except an additional labeling step. |
