Preparation of chitosan-based nanoparticles for efficient pro-oxidant cancer therapy |
초록 |
Development of biocompatible nanoparticles with low toxicity and enhanced bioavailability was of great interest for anticancer drug delivery.Chitosan(CS) is a cationic polysaccharide and obtained by deacetylation of chitin.CS has high biocompatibility and antimicrobial properties and thus has been used as a safe drug carrier.It also has features that can control the drug release in a pH-dependent manner.Fucoidan(FUC), a sulfated polysaccharide that can be extracted from brown algae, has been shown to have antioxidant and anti-inflammatory effects.Piperlongumine(PL) is a pro-oxidant agent which mediates apoptosis by increased oxidative stress in cancer cells.In this study, biocompatible chitosan and FUC were used to prepare nanoparticles encapsulating hydrophobic PL.The nanoparticles via ionic gelation of chitosan and FUC stably encapsulated PL and increased its bioavailability.The PL-loaded nanoparticles efficiently killed human cancer cells via increased oxidative stress in the cells. |
