Mammalian cells require more than two biological factors for proliferation or differentiation mostly. In order to optimize the effects of factors, they should be used at appropriate amounts for specific periods. Thus it is very important to control the release of each factor independently. We fabricated electrospun nanofiber scaffolds combined with PEG hydrogel for controlled release of bFGF and BMP2 individually, and this system can induce differentiation of hMSC to osteoblast efficiently. The electrospun fibers and hydrogels were used as reservoir of growth factors respectively. Because each has distinguished release profile, the amounts and periods of release were regulated by location of growth factors. hMSCs were cultured on the scaffolds, and usefulness was showed from differentiation of hMSCs. This independent controlled release system has a great potential for the bone tissue engineering. |
