A Novel DNA Carrier using Vinylimidazole copolymer |

To prepare a new cationic polymer, poly(1-vinylimidazole-co-4-vinylimidazole) (P14V) by copolymerization of 1-vinylimidazole (1VIm) and 4-vinylimidazole (4VIm) as a gene carrier. 4VIm was prepared by decarboxylation of anhydrous urocanic acid. P14V was synthesized by copolymerization of 1VIm and 4VIm with azobis(isobutyronitrile) (AIBN) as an initiator at 60℃ for 24 h under nitrogen. The prepared P14V was confirmed with H1-NMR. The complex formation between P14V and DNA was confirmed using agarose gel electrophoresis. And the DNA in P14V/DNA complex can be protected even at higher concentration of the nuclease than the physiological condition. P14V showed less cytotoxicity than polyethylenimine (PEI). The gene expression was occurred in HeLa and MC3T3-E1 cells. The P14V showed good transfection efficiency in in vitro. As a results P14V is expected to use as gene carrier for gene therapy.
