Biocompatible Polymer Coated Gold Nanoparticle for CT/Optical Dual Imaging |

Gold nanoparticles (AuNPs) have attracted much attention as their intrinsic properties. However, AuNPs cannot be used in the biomedical field because of the instability in physiological condition. To improve stability of the AuNPs in physiological condition, AuNPs were coated with biocompatible Glycol Chitosan (GC). GC is one of the excellent candidates to introduce stability of AuNPs. In this study, we described strategies for stabilized AuNPs using GC and verified their potential for biomedical application. GC Coated AuNPs (GC-AuNP) were intravenously tested as a Computed Tomography (CT) contrast agent in vivo mice tumor (SCC-7) model. And the AuNPs were conjugated with Cy5.5-MMP (Matrix MetalloProtease) specific substrate, which can be used as a MMP specific fluorescence imaging probe for tumor progression. The Cy5.5-MMP conjugated GC-AuNPs were intravenously injected in mice bearing SCC-7 tumors for tumor detection. That showed their accumulation in tumor by EPR effect. |
