Expression of DOI synthase (kanA) in Streptomyces lividans TK24 and Comparison with E. coli Counterpart |

A cluster for the kanamycin (Km) biosynthesis has been isolated from the genomic library of S. kanamyceticus ATCC 12853. Sequencing of 47 kb region or chromosomal DNA revealed 40 open reading frames (ORFs) including Km biosynthetic, regulatory and resistance genes [2]. For expressing actinomycetes genes in Streptomyces spp, an expression vector (pIBR25) was constructed and used to express a gene encoding 2-deoxy-scyllo-inosose synthase (kanA) in S. lividans TK24 from the Km biosynthetic gene cluster. The activity of kanA was dependent on the culture period of the transformants reaching maximum at 72 h. The application of sorbitol and betaine in the expression medium improved the formation of soluble KanA as well as its activity when it was expressed in Escherichia coli. The results provide a valuable tool for expressing aminoglycoside-aminocyclitols (AmAcs) biosynthetic genes in Streptomyces spp. |
