제목 | Biosynthesis of light-harvesting phycobiliproteins in a cell-free system |
초록 | Microalgae are considered as a potential workhorse to produce biofuels and other chemicals of industrial implications. However, unlocking the power of microalgae should require extensive engineering, which is limited by the paucity of efficient tools for genetic manipulation of these organisms. In this study, we probe the possibility of employing a cell-free synthesis system as a breadboard for studying the metabolic pathways of microalgae. As an exemplary model, we chose the biosynthetic pathway of phycoerythrin, one of the light-harvesting proteins of microalgae, a red pigment-protein complex. A cell-free synthesis system derived from Escherichia coli was programmed with PCR-amplified genes of the enzymes involved in phycoerythrin synthesis. During the incubation of the cell-free synthesis reaction mixture, we were able to confirm the activities of these enzymes determined by the changes in absorbance. |
