Synthetic gene circuits and switches for cell-based and cell-free applications |

Synthetic gene circuits and switches are increasingly used to control gene expression in various cellular and cell-free applications. The remaining challenges in building synthetic gene circuits and switches for more practical applications are to minimize cellular burden to maintain the genetic circuits but to provide simple biocontainment options. First, we use empirical design and iterative construction to build single-copy, bistable toggle switches with reduced metabolic load. The theory to create the single-copy circuit has been implemented to develop biocontainment system that couples environmental sensing with circuit-based control of cell viability. To prevent release of engineered microbes into open environmental applications, we also suggest cell-free in vitro translation and transcription as a means to produce and deliver synthetic functions. Overall, the given approaches that resolves important practical limitations can be applied to develop future synthetic devices. |
