Polyamidoamine(PAMAM) dendrimer is polymer consisting of peptide bond between ethylenediamine and methyl acrylate and has high biocompatibility and biodegradability. However, It has relatively poor transgene expression capacity than Polyethyleneimine(PEI). In order to solve such problem, we introduced Nuclear Localization Signal(NLS) Peptide to PAMAM dendrimer. NLS peptide is factor assisting entry of external gene into the Nuclear Pore Complex(NPC) for active transport. We used pentapeptide originated from adenovirus e1a, which is KRPRP. We synthesized two type PAMAM – KRPRP and PRPRK(arginine is N-terminus) so as to verify difference of induced functional status from inversion of N-terminus and performed experiment in several cell line. PAMAM-PRPRK indicated similar transfection efficiency corresponding to PEI in HEK 293 cell line and also shows comparatively lower cytotoxicity than PEI. |
