Despite high silencing efficiency and on-target specificity, the clinical translation of siRNA has been hindered by its inherent features: poor intracellular delivery, limited blood stability, and unwanted off-targeting effects. For overcoming these hindrances, I suggested polymerized siRNA (poly-siRNA), which can produce highly condensed and homogenous complexes with thiolated natural polymers, as a new concept for local and systemic siRNA delivery. In my presentation, I will briefly review the history of poly-siRNA-based delivery system. Then, I will discuss several therapeutic applications of poly-siRNA in different diseases. |
