제목 In Vitro Evaluation of Release Profiles of Fentanyl Loaded-Poly(lactide-co-glycolide) Microspheres
초록 Fentanyl is a potent synthetic opiate and commonly used for the treatment of severe pain in cancer patients. However, it shows a very rapid onset of action in vivo application. In this study, we developed several kinds of fentanyl loaded microspheres (FMS) for sustained release of fentanyl. PLGA oligomer, molecular weight 8000 g/mole, was used to fabricate fentanyl loaded microspheres (FMS) for sustained release of fentanyl. FMS were prepared by an emulsion solvent-evaporation method. The influence of several preparation parameters such as initial drug loading, PLGA concentrations, emulsifier concentrations, and oil phase volume have been investigated on the fentanyl release profiles. The size of FMS was very fine with 20∼90 μm which is considered to be suitable for i.m. injection. We also developed a rapid and sensitive determination method by employing gas chromatography with nitrogen specific detection (GC-NPD) system. Generally, the drug showed the biphasic release patterns, with an initial diffusion followed by a lag period before the onset of the degradation phase, but there were no lag time in our system. Fentanyl was slowly released from FMS over 10 days in vitro. From the results, FMS may be a good formulation to deliver the anesthesia for the treatment of chronic pain.
