A variety of vaccine delivery systems have been developed for easy administration, cost-effective, and efficient immunization for patient compliance. Among them, transdermal vaccine delivery systems appeared to be attractive due to the strong immune responses in Langerhans and dendritic cells of dermal tissues. In this work, we developed smart transdermal vaccine delivery systems using hyaluronic acid (HA) derivatives. As a model system, HA-ovalbumin (HA-OVA) conjugates were synthesized and characterized by GPC and ELISA. After transdermal delivery, intravital microscopic imaging clearly visualized the penetration of HA-OVA-rhodamine B conjugates through the skin into deep dermal tissues. Furthermore, the HA-OVA conjugates significantly enhanced the immune responses showing a 15 times higher anti-OVA IgG concentration in 4 weeks than the control of OVA. Taken together, we could confirm the feasibility of HA-based transdermal vaccine delivery systems for further clinical applications. |
