Collagen/Chitosan-nanofiber hydrogel as a carrier for growth factor delivery |

Growth factor (GF) stimulates the proliferation and the differentiation of hMSCs. Controlled release of GF, such as BMPs, should maintain biological activity of GF for bone tissue engineering applications. Poor mechanical property of collagen (Col) hydrogel caused significant shrinkage during 3D culture. In order to overcome these problems, BMPs were introduced in chitosan nanofibers (CS-NFs) by using LBL methods. Col hydrogels were reinforced with BMP-loaded-CS-NFs (BCNFs). Elastic modulus of hydrogel, distribution of BCNFs in hydrogel, and release kinetics of BMPs were measured using UTM, confocal microscopy, and ELISA, respectively. Shrinkage behaviors of hydrogels on culture were measured as function of times. Mechanical properties of Col/CS-NFs hydrogel were increased with increase in CS-NFs concentration. The proliferation and osteogenesis of hMSCs in BMP-2 delivered Col/CS-NFs hydrogel have been evaluated. |
