Determination of Platelet Functional State Using Polydiacetylene based Biosensor |

Clinical evaluation of the functional state of platelets is mainly dependent presently on patient medical histories. In developing a simple test for platelet assessment the biosensing literature was investigated to find a platform for detection of activated platelets. Direct detection of platelet allows the determination of the extent of platelet activation under most conditions including contact activation in extracorporeal circulations, antiplatelet drug effects and hereditary platelet diseases. We have developed a simple, rapid, and sensitive whole blood method to measure platelet function. PDA changes shape and color in response to molecular stress. Molecular stress occurs when activated platelets bind to a specific platelet antibody-linked PDA. The PDA method : A whole blood sample is stimulated with kaolin, collagen or ADP. The platelets aggregate, binding fibrinogen, and the color of the PDA changes based on the amount of activated fibrinogen/ bound to platelet complexes. |
