We demonstrate highly sensitive and selective potassium detection against excess sodium ions, by modulating the interaction between G-quadruplex-forming molecular beacon aptamer (MBA) and a cationic conjugated polyelectrolyte (CPE). High binding specificity of MBA for potassium ions offers selectivity. Conformational changes of MBA by complexation with CPE and the signal quenching or amplification via fluorescence resonance energy transfer in the presence and absence of potassium ions, provide the remarkable sensitivity with the lowest detection limit of ~1.5 nM among reported so far. Our scheme reported herein can be readily applicable to sensing other kinds of G-rich aptamer-binding chemicals and biomolecules. |
