PLGA is one of the most widely used polymers in pharmaceutical applications. Extensive studies have been conducted to elucidate the effects of such parameters as molecular weight, polydispersity, and monomer composition on the properties of PLGA. However, studies dealing with the effect of monomer sequence distribution have been sparse. Herein, we present a scalable semi-batch copolymerization strategy that enables the production of statistically sequence-controlled PLGA through control of the rate of comonomer addition. Using this feed rate-controlled semi-batch copolymerization method, a series of PEG-PLGA samples having an identical molecular weight and monomer composition but different sequence distributions were prepared. The measured physiochemical properties significantly varied among the samples, demonstrating that the implementation of comonomer sequence control only at the statistical level still significantly influences the properties of the copolymer products. |
