Rheumatoid arthritis (RA) is a chronic inflammatory disease caused by the inflammation of synovial membrane leading to articular cartilage destruction. In this work, tyramine modified hyaluronic acid (HATyr) conjugate was synthesized. Then, HATyr hydrogels were prepared to encapsulate dexamethasone (DMT) and its conjugate with monoclonal antibody (mAb). Intra-articular injection of HATyr hydrogels with DMT showed an excellent therapeutic effect on RA with reduced interlukine-6 (IL-6), prostaglandin E2 and four types of cytokine levels in collagen-induced arthritis animal models. Histological analysis also confirmed the therapeutic effect of injectable HATyr hydrogels with DMT. Currently, the therapeutic effect of HATyr hydrogels encapsulating DMTmAb conjugates are under investigation for further development. Taken together, the injectable HATyr hydrogels were thought to be developed as a therapeutically effective drug carrier for the treatment of RA. |
