제목 Doxorubicin loaded biodegradable poly(ethylene glycol) methyl ether acrylate-b-poly(L-lysine)-b-poly(L-histidine) chimeric micelles for pH-sensitive intracellular anticancer delivery
초록 To produce an endosomal pH responsive delivery vehicle, we have synthesized a series of polypeptide containing triblock copolymer, poly(ethylene glycol) methyl ether acrylate-block-poly(L-lysine)-block-poly(L-histidine) (p(PEGA)30-b-p(Lys)25-b-p(His)n) (n = 25, 50, 75, 100) via the combination of reversible addition fragmentation transfer polymerization, ring opening polymerization of α-amino acid N-carboxyanhydrides and azide-alkyne click cycloaddition reactions. An anticancer drug, doxorubicin (Dox), was effectively encapsulated in the micelles fabricated by self-assembly process and delivered under different pH conditions. The polymer exhibited very low in vitro cytotoxicity which demonstrated efficient pH sensitive intracellular delivery of Dox into CT 26 cell lines.
