Biocompatible polymers-coated magnetic nanoparticles as ultra sensitivecontrast agent. |
초록 |
Biocompatible polymer nanoparticles containing a cluster of manganese-doped superparamagnetic nanoparticles(Mn-SPIONs) were simply fabricated by ionic assembly between poly-γ-glutamic acid(γ-PGA) and chitosan. Mn-SPIONs coated with hydrocarbon molecules were synthesized via thermal decomposition method. Mn-SPIONs were transferred into aqueous phase via ligand exchange with γ-PGA. Polymer nanoparticles were formed by ionic assembly between negative charged γ-PGA coated Mn-SPIONs and positive charged chitosan, and the size of particles was 80~150nm, confirmed by SEM, TEM and DLS. Polymer nanoparticles presented high value of MRI relaxivity, 507(Mn+Fe) mM-1S-1, which is much higher than that of conventional iron oxide-based MRI contrast agents, due to their synergic magnetism effect. These nanoparticles were easily taken up by RAW 264.7 cells, and it was confirmed by prussian blue staining and a strong darkening effect of T2-based MR images of Mn-SPIONs labeled RAW 264.7 cells. |
