pH – responsive organic nano-indicator tailored by pseudo metal for trafficking endolysosomal compartment |

Intracellular pH plays a crucial role in cell biology since proton gradients underlie the function of organelles such as endosome, lysosomes and mitochondria. Therefore, observation of proton gradients in cell components is essential. One promising approach is the assessment of nanoparticle based on polyaniline (PANI) that exhibited convertible transition state by proton which had the feasibility as observer of intracellular pH. Here, we synthesized silica coated PANI nanoparticles (PSIsNIs) as indicator. First, hetero structure of Fe3O4/MnO (IsNIs) was synthesized and coated with silica (SIsNIs). Subsequently, PSIsNIs were formed by IsNIs as oxidant and shifting agent of transition state from pH3 to pH6. Finally, Cy3 and Cy7 as pH insensitive dye was adsorbed and PEGylated in PSIsNIs, respectively (FPSIsNIs). To assess the potential of FPSIsNIs as indicator, we investigated morphology, optical properties, and we evaluated quenching ability according to proton gradient in vitro. |
