Transdermal Hyaluronic Acid – Vaccine Conjugates for Needle-free Facile Immunization |

Transdermal vaccine delivery has been regarded as an efficient way for immunization due to the strong immune responses of Langerhans cells and dendritic cells (DCs) in dermal tissues. In this work, we developed needle-free transdermal vaccine delivery systems by the conjugation of vaccine molecules with hyaluronic acid (HA). As a model system, HA-ovalbumin (HA – OVA ) conjugates were synthesized by the site-specific conjugation of HA-aldehyde with N-terminal amine group of OVA. The successful synthesis of HA – OVA conjugate was confirmed by GPC, circular dichroism and ELISA. Intravital microscopy clearly visualized the skin penetration of HA – OVA conjugates into deep dermal tissues. After transdermal delivery, HA – OVA conjugates exhibited strong immune responses due to the self-adjuvanting HA and the conjugated OVA. The significantly enhanced immunization was confirmed by elevated serum anti-OVA IgG concentration and enhanced cytotoxic T lymphocyte responses. |
