The fields of non-viral gene therapy are rapidly gaining increased interest owing to the potential advantages of these non-viral carriers from view points of their safety, production, and versatility. In this study, reaction of the polyethylenimine(PEI) to a poloxamer diacrylate as a nonionic water-soluble polymer was performed to get a non-toxic and biodegradable gene carrier. The molecular weight and degradation of the PEI-co-Poloxamer (PEI-POL) were determined using viscosity measurement. The PEI-POL/DNA complex was confirmed using agarose gel electrophoresis and DNase I experiment. The sizes and zeta potentials of the complex were measured by dynamic light scattering. The cytotoxicity of PEI-POL in HepG2 and HeLa cells was evaluated. The PEI-POL/DNA complex was transfected into HepG2 and HeLa cells. |
