Mannose Receptor-Mediated Gene Delivery into Antigen Presenting Cells |

The modulation of immunity, in particular of T cells, is an important goal in treatment and prevention of viral infections, cancer and autoimmune disease. At the beginning of every cellular immune reaction, antigens have to be presented to T cells in order to activate them and drive them into clonal expansion. This is done by antigen presenting cells (APCs), and the most powerful of which is the dendritic cells (DCs). A fairly new approach in vaccine research is the development of antigen-encoding plasmid DNA as immunogen. To deliver DNA vaccine into APCs, non-viral vectors are strongly in need as safer alternatives to viral vectors. In this study, to induce the receptor-mediated endocytosis into APCs having mannose receptors, mannosylated chitosan(MC) was prepared as a gene carrier. Chitosan was coupled with mannopyranosylphenylisothiocyanate bearing mannose group as a specific ligand and the composition of MC was determined by NMR spectroscopy. Complexes were induced to self-assembly and complex formation and DNAse I resistance were confirmed by electrophoresis. Particle sizes and morphology of the complexes were determined using TEM. Cytotoxicity and transfection efficiency of MC were investigated in comparison with chitosan itself. |
