In vascular tissue engineering, structural similarity and biological function of the scaffolds are very important to develop vascular graft resembling native blood vessel. To develop artificial blood vessel, we fabricated electrospun fibrous meshes with nanofibrous layer and microfibrous layer, and then each layers were functionalized with acrylic acid by gamma-ray irradiation. To enhance biological function of the scaffolds, we further immobilized VEGF and gelatin as bioactive molecules regulating cellular responses, and following characteristics of the modified scaffolds were analyzed with SEM, FT-IR, ELISA, and fluorescamine assay. In cell experiments, our functional meshes successfully regulated behaviors of HUVEC and SMC, consequently it can be alternative scaffolds for vascular tissue engineering. |
