Polydiacetylene(PDA) liposome based detection of Alzheimer's disease biomarker |

The research for highly selective and sensitive label-free biosensor has been widely investigated with a perspective change of Alzheimer’s disease (AD) from cure to management and prevention. Here, we fabricated PDA vesicle-based biosensor for detection small ubiquitin-related modifier-1(SUMO-1) protein, as an AD biomarker. PDA vesicles comprised of PDA monomer and phospholipid were inter-connected with an linker to endow with flexibility and adhesiveness on the substrate. In addition, secondary interaction based on the polyclonal antibody was introduced to amplify the signal of PDA liposome sensor. Detection and quantification of SUMO-1 protein can be measured by the intensity of fluorescence triggered by the ligand-receptor interaction of PDA liposome. The increased fluorescence intensity in designed PDA liposomes showed that sensitivity of PDA biosensor was improved. The strategies used in the system can be applicable to PDA liposome-based sensors for various diseases models. |
