Enzyme-mediated injectable hydrogel for tissue engineering and drug delivery |

Recently, injectable hydrogel systems using end-group-chemical reaction have been widely investigated. In particular, enzymatic cross-linking systems using horseradish peroxidase (HRP) and hydrogen peroxide (H2O2) have some advantages, such as biocompatible and fast reaction in mild condition. In this study, injectable hydrogels composed of Tetronic-tyramine (Tet-TA), gelatin-hydroxyphenylacetic acid (GHPA) and heparin-tyramine (Hep-TA) conjugates were developed. The conjugates were synthesized by a common carbodiimide/active ester-mediated coupling reaction. The hydrogels were rapidly formed under physiological condition in presence of HRP and H2O2. Their physico-chemical properties were characterized and in vitro cell culture was investigated using osteoblast. In addition, in vitro growth factor release test was carried out. Obtained results demonstrated that the enzyme-mediated injectable hydrogel is a promising injectable material for tissue engineering and drug delivery. |
