Screening of bioreducible CBA-amine polymers for the effective intracellular delivery of plasmid DNA |

Here, we describe novel bioreducible cystamine bisacrylamide(CBA)-amine polymers as new plasmid gene delivery carriers. Reducible disulfide bonds in the backbone of CBA-amine polymer can be readily break down to release the genetic material into cytosol by glutathione(GSH) in cytosol. We have synthesized and screened a library of bioreducible CBA-amine polymers and characterized their gene transfection efficiency in vitro. We have identified CBA-amine polymers with excellent transfection efficiency by inducing GFP expression on MEF cells. The complex formation between the polymers and pDNA was confirmed by gel retardation assay and their size was analyzed, in which, they were in the range of 200~500 nm by DLS measurement. We also found that the viability of transfected cells was significantly higher than conventional gene carrier such as lipofectamine. These results indicated that CBA-amines polymers have significant potential to use as promising gene delivery carriers for direct cell reprogramming. |
