Transplantation of ex vivo cultured cells is one of the most emerging research fields in tissue engineering. In particular, the cell delivery system is of critical importance in this approach. Conventional cell delivery system involves direct cell injection or scaffold-based system. However, these systems have often reported several problems such as poor engraftment efficiency, low cell survival, rejection or toxicity of implanted biomaterials. Alternatively, transfer printing of cells has been considered to transplant tissue-like construct, often referred as cell sheet with maintaining intact cell-cell junction. We developed cell-interactive and temperature responsive hydrogels as a novel substrate to culture confluent cell layers, which could be successfully transferred to new substrates including tissue by reduction in temperature. Furthermore, we confirmed a therapeutic potential of our system using in vivo tissue defect models. |
