제목 NIR-responsive Fluorescent Carbon Dot Platform for Targeting Combined Thermotherapy and Mitochondrial Dysfunction in Cancer Treatment
초록 In this work, we synthesized a core shell platform based on fluorescent carbon dot (FNP) owning photothermal properties with the attachment of the glycolysis inhibitor-3-bromopyruvate (BP) and delocalization of positively charged lipophilic triphenylphosphonium (TPP) and folic acid (FA) for mitochondrial and tumor targeting. FNP was synthesized from the carbonization of polyethylene glycol (PEG) grafted poly (dimethyl aminoethyl methacrylate)-q-catechol to be used as core. The shell was constructed by the conjugation of boronic acid (BA), BP, TPP, FA and a photothermal agent – IR825 with (PEG-g-PDMA) backbone, then crosslinked with FNP to produce F-FNP. The NIR-responsive F-FNP matrix showed its ability to target and block mitochondrial function and antitumor properties via photothermal therapy. This core-shell antitumor platform offers a new pathway to efficiently target and kill cancer cells. This work was supported by Korea National University of Transportation in 2018.
