Development of Multifunctional Gold Nanoparticles for Cancer-specific CT Imaging and therapy |

Gold induces a strong X-ray attenuation, and its nano-sized particles have unique physical, chemical and biological properties, which make them an ideal candidate for CT contrast agents. Here we synthesized multifunctional gold nanoparticles (MFGNPs) as a CT imaging agent. Their surface was coated with PEG for enhancing blood circulation time. Branched PEI were also conjugated for the immobilization of targeting ligand and therapeupic materials. Physicochemical properties of MFGNPs were characterized by TEM, DLS, Zeta-potential and TGA. Thereafter, we investigated endocytic pathway of MFGNPs in CT-26 cell line in vitro, and further examined in vivo biodistribution of MFGNPs and their cancer targeting effect in tumor-xenografted mice. In conclusion, we confirmed intracellular uptake in vitro and in vivo. MFGNPs could be utilized as a molecular probe for early diagnosis of cancer or cell labeling. In addition, it will be easily developed with introduction of other bioactive agents. |
