Osteogenic differentiation of bone marrow derived human mesenchymal stem cells(hMSCs) on electrospun poly(L-lactide-co-ε-caprolactone)(PLCL) and gelatin composite nanofiber scaffolds |

In tissue engineering, it is important to develop scaffolds that can be functional to control morphogenic response of progenitor cells and to direct desirable regeneration. In this study, we fabricated three-dimensional nanofibrous scaffolds by electrospinning polymer blends of poly(L-lactide-co-ε-caprolactone)(PLCL) and gelatin to modulate differentiation of human mesenchymal stem cells (hMSCs). We hypothesized that the combination of natural and synthetic polymers may provide an ideal substrate for hMSCs culture. hMSCs were seeded on four types of scaffold and induced to differentiate along osteogenic lineages. We examined gene expression of osteogenic markers, calcium deposition, and alkaline phospatase activity. Cells seeded on PLCL/gelatin blend scaffold showed higher level of ALP activity and calcium deposition than those seeded on PLCL scaffold. These results suggest that PLCL/gelatin blend nano-fibrous scaffolds may provide biomimetic ECM environment for tissue regeneration. |
