We have developed several kinds of vector systems using polymeric nano-materials for efficient delivery of siRNA. To immobilize siRNA to polymers and nanoparticles, the selection of optimal conjugation strategies including cross-linkers, cleavability, and conjugation sites is a crucial step for proper RNAi processing after intracellular uptake. Various conjugation strategies to incorporate siRNA and imaging agents onto polymer-coated inorganic nanomaterials will be paid attention for biological applications in vitro and in vivo. Particularly, using one of optimal conjugation method, siRNA were immobilized onto iron oxide nanoparticles coated with biocompatible polymers, which could exhibit efficient siRNA delivery as well as MR imaging for cancer cells. These multifunctional polymeric nanomaterials for biomedical imaging and therapy might have powerful potential not only for treating incurable diseases but also for monitoring prognosis of cancers. |
