Development of polyamine-based nanoparticles for small interfering RNA delivery to hepatic cells |

Small interfering RNA (siRNA) can mediate a powerful gene-silencing for treatment of varied diseases. Polyamines could be used to synthesize siRNA delivery materials due to their cationic characteristics. Here we developed polyamine-based lipid-like nanoparticles for siRNA delivery to human hepatic cells. Alkyl-epoxides were conjugated to several polyamines through ring-opening reactions. In a reporter siRNA delivery experiment, among polyamines spermine was found to be effective for siRNA delivery to human hepatic cells. When spermine-based nanoparticles were tested for delivery of siRNA targeting a host enzyme, PRK2, which is required for hepatitis C virus replication, they mediated significant silencing of the target enzyme expression in human hepatic cells.
