제목 Reduction-responsive crosslinked polyelectrolyte complex formulation for siRNA delivery.
초록 Gene regulation using siRNAs has been recently recognized as a potential therapeutic tool due to their sequence-specific action by an extremely low intracellular concentrations. Despite enormous therapeutic potentials of siRNA, their clinical applications have been limited because of the lack of efficient delivery systems. For developing efficient siRNA delivery strategy, herein we synthesized bile acid-modified cationic polymers, polyethyleneimine-deoxycholic acid (PEI-DA), and their siRNA polyelectrolyte complexes were further stabilized through post-condensation disulfide crosslinking. The disulfide-crosslinked PEI-DA/siRNA complexes maintained their stable nanostructures under extracellular low reducing conditions and promptly released siRNA in response to redox potential. The stabilized PEI-DA/siRNA complexes also exhibited efficient transfection efficiency due to the enhanced membrane permeability resulted from amphipatic bile acid (DA).
