제목 The Inflammatory Reaction Reduction to Fibrin/PLGA Hybrid Scaffold ; In Vitro Test
초록 PLGA(lactide-co-glycolic acid) (PLGA) has been widely applied to tissue engineering as a good biocompatible material. However, their acidic degradation products can decrease the pH in the surrounding tissue, which result in local inflammatory reaction. We hypothesized that the cells neighboring the PLGA implant might have an inflammatory response which could be reduced by impregnating fibrin into the PLGA. Fibrin/PLGA scaffolds were prepared by solvent casting/salt leaching method. In vitro, cell viability and proliferation analyzed by MTT .The release profiles of TNF-α were assayed using ELISA. In this study, we assessed the effect of fibrin for reducing inflammatory response of PLGA through several biocompatibility tests in comparison with PLGA. Fibrin/PLGA scaffolds showed higher cell viability and lower production of inflammatory cytokine than PLGA scaffold. This research was supported by SCRC(SC4110)and WCU(R31-20029)
