제목 The translocation of elastin-like polypeptide for gene delivery
초록 Efficient gene delivery would significantly impact in clinical therapies, diagnostics and basic science research. For non-viral vector translocation, biomaterials are a critical component for the development of efficient delivery systems. In the presentation, elastin-like polypeptide (ELP) was used for a vehicle for specific cellular uptake because of its inverse transition temperature (Tt) and biocompatibility. Thermoresponsive elastin-like polypeptide (ELP) was synthesized by recombinant DNA technique. Inverse transition temperature of stimuli-responsive bioactive ELP was characterized using UV spectrometer. The phase transition resulted in changes of binding affinity for plasmid DNA. For improving cellular uptake efficiency, ELP fused to cell penetrating peptide (CPP) was constructed. Cellular uptake efficiency was investigated through expression level of green fluorescent protein.
