We present the ultrasensitive biosensors using dual antibody-linked immunoassay based on magnetic nanoparticles (MNPs) and fluorescent dye-loaded polymersomes (Psomes). The MNPs are capable of separating and purifying the target materials and Psomes encapsulates the dye to amplify the signal. In the presence of target biomarker, MNPs and Psomes, which are surface modified with different antibodies, bind to both sides of the biomarker to form a sandwich complex. We then confirmed the presence of biomarker after lysis of Psomes that release the encapsulated dyes by providing one-to-multi signals. We established MNP-Psomes based immunoassay to diagnosis diseases with ultra-sensitivity. Further, in order to confirm whether or not the protein can be selectively separated, a biotin-avidin precedent experiment was performed. In addition, it can be applied to various disease depending on the target biomarker. |
