Ladder Library Synthesis for Serine Kinase Substrate Screening using One-Bead One-Compound (OBOC) method |
| 초록 |
Determinination of the phosphorylation site of proteins by kinases is essential for the study of regulation of cell functions. We have developed a method of the solid phase ladder peptide synthesis for preparing one-bead one-compound (OBOC) combinatorial peptide library that is efficient for the screening the substrate specificity. The the peptide sequence of serine kinase substrate is determined by using MALDI-TOF MS, which is used to measure mass differences between the adjacent members of the N-terminated peptide products. |
