Preparation of core-shell type MBHA resin and its application to Solid-Phase Peptide Synthesis |

MBHA(4-methylbenzhydrylamine) resin has been widely used for the synthesis of peptide amides and its industrial demand as a solid support for the production of peptide drugs is increasing. We prepared MBHA resin by benzotriazole-catalyzed amidoalkylation. First, formamido PS was synthesized from 1% DVB-PS using bis-formamide and benzotriazole. By removing formamido group, MBHA resin was obtained. The core-shell type MBHA was also constructed by partial hydrolysis of the formamido groups. It was analyzed by confocal microscopy and its synthetic performance in SPPS was evaluated. |
