Cytophilic surfaces can be achieved by physical or chemical modification of surfaces with cell-signal molecules. Using the specific binding affinity of heparin, the surface properties of heparin-based hydrogel can be modulated easily by physisorption of cell-adhesive proteins. For example, Fibrinogen was adsorbed efficiently onto the heparin hydrogel via heparin affinity by simple incubation with fibronogen solution. The physisorption of fibrinogen was so strong and relatively stable and fibroblasts attached and spread well on the fibrinogen bound hydrogel. Cell proliferation was also remarkably enhanced, similar to that on tissue culture plate, and the optimal cell proliferation was observed with the incubation at 1 mg/mL of fibrinogen. In contrast, serum adsorbed hydrogel did not change the cell-adhesive properties of the hydrogel. Therefore, it is possible to selectively change the cytophilicity of the hydrogel and this approach can be utilized in various bio applications. |
