제목 Molecular Design and Synthesis of pH-sensitive Polymeric Sensor For Intramolecular Fluorescence Resonance Energy Transfer
초록 Molecular imaging in tumor biology is becoming one of the most active scientific and clinical research areas. Especially, optical imaging based on fluorescence resonance energy transfer (FRET) phenomena has emerged as an important technique to describe clinical targets in vivo. The goal of this study is the development of polymeric biosensor which emits specific fluorescence upon responding to lowering pH. In this work, sulfadimethoxine is chosen for pH sensitive moiety. Sulfadimethoxine is conjugated with vinyl benzyl chloride to prepare styrene type monomer. pH sensitive polymeric backbone is prepared via atom transfer radical polymerization (ATRP) of styrene type sulfadimethoxine monomer. This polymeric backbone could be reversibly changed in end-to-end distance, which comes from the unique properties of pH sensitive sulfadimethoxine. For FRET study, pyrene and coumarin are selected as FRET donor and FRET acceptor, respectively. Pyrene and coumarin are positioned at both ends of polymeric backbone from ATRP of styrene type sulfadimethoxine monomer using pyrene-containing initiator, followed by chain end functionaliztion by using coumarin derivative. FRET from pyrene to coumarin as a function of pH change will be studied.
