Binderless granulation of cohesive fine powder by using a spouted bed and its application to particle design for dry powder inhalation
To develop a new process for the granulation of cohesive fine powder, the granulation without binder was investigated by using a spouted bed. The produced granules in this method were almost spherical. The diameters of granules were not affected,by the granulation conditions, such as the fluidization gas velocity and taper angle of bottom of apparatus. However, it was found that the diameter of granule could be controlled by changing the initial sieving condition of raw powder. Although the compressive strength of produced granules was too small to be measured with GRANO particle hardness tester, the granules were hard enough for the handling. This binderless granulation method by a spouted bed applied for the dry powder inhaler (DPI). The dry powder inhaler has been attracted attention as the cure for bronchial asthma. The medicine was inhaled to bronchia by a patient's breathing through DPI equipment without using chlorofluocarbon. Therefore, the DPI required good flowability and dispersibility, and should be dispersed easily about 0.5 to 7 mu m in aerodynamic particle diameter at the inhalation. Actual medicines were used for the granulation material in the granulation experiment. The dispersibility of granule was measured by the cascade impactor. As the result, the dispersion was satisfactory for DPI and the produced granules by the granulation method had good dispersion quality than that by the conventional granulation method.
