Halobacterium Halobium에서 Purple Membrane을 재구성시킨 Vesicle의 마취제에 따른 부피와 에너지변화

Purple membrane과 몇가지 지질의 현탁액에서 1-펜탄올, 에틸알콜, 할로테인 등의 마취제를 첨가한 혼합물의 잉여부피는 dilatometer를 사용하여 25℃에서 측정하였다. 일반적 마취력은 지질 용해도와 관련성이 있다. Halobacterium Halobium의 purple membrane에서 막 단백질에 의한 vesicle의 변성은 광화학반응인 상전이와 부피변화를 연구하였다. 혼합물의 잉여부피는 에틸알콜에서 음의 부피를 나타내지만 할로테인에서 양의 부피를 확인하였다. 이 결과는 Mill의 가정에 일치한다. 또한 purple membrane이 재구성된 vesicle에 혼합한 마취제의 thermogram으로부터 상전이온도를 연구하였다. 마취제와 purple membrane이 재구성된 vesicle 계가 마취제와 purple membrane 계와 비교해서 상전이가 더 높게 나타났다.
The excess volumes of mixture of 1-pentanol, ethyl alcohol and halothane in purple membrane and in suspensions of lipids have been determined at 25℃ using a excess volume dilatometer. The potency of general anesthetics has long been known to correlate with lipid solubility. Denaturations of the vesicle in the purple membrane of Halobacteriun Halobium were studied by volume changes and photochemical reaction differential scanning calorimetry. The excess volumes of mixture in ethyl alcohol were all found to be negative, whereas they were positive in halothane. The result was conformed to Miller''s supposition. Thermograms of purple membrane vesicle systems incorporated with anesthetics have been studied. Phase transition temperature of vesicles incoporated with anesthetic and purple membrane was higher than that of the mixture of anesthetic and purple membrane.
